KARAKTERISASI TINGKAT VIRULENSI BAKTERI Vibrio splendidus PADA KERANG MANILA (Ruditapes philippinarum) MENGGUNAKAN TES IN VITRO

Abstract

Abstrak - Vibrio splendidus merupakan penyebab terjadinya Brown Ring Disease (BRD) pada kerang manila, Ruditapes philippinarum. Bakteri yang diujikan didapat dari Laboratoire de Biotechnologie et Chimie Marines (LBCM), Prancis. Karakterisasi bakteri dilakukan dengan menggunakan tes in vitro. Penelitian ini dilakukan untuk mengevaluasi tingkat virulensi dai V. splendidus dengan menggunakan serangkaian tes in vitro pada kerang manila. Pada tes yang dilakukan, kami menemukan filopod pada hemosit kerang hilang, yang menandakan adanya sitotoksisitas. Untuk menghitungnya, beberapa tes flow cytometry seperti uji aderensi, fagositosis, dan reactive oxygen species (ROS) tes dilakukan. Dua jenis bakteri Vibrio tapetis digunakan untuk mebandingkan aktivitas keduanya. Hasil dari penelitian ini ditemukan bahwa aktivitas Vibrio splendidus lebih rendah dari Vibrio tapetis dan kontrol negatif.

Abstract - Vibrio splendidus is the causative agent of Brown Ring Disease (BRD). This bacterium affects the manila clam, Ruditapes philippinarum. A strain of V. splendidus was obtained from LBCM. The bacterium was characterized its ability to form biofilms in vitro. This study was designed to evaluate the virulence of V. splendidus strain using in vitro assays to the R. philippinarum. In in vitro assay, we found clam hemocytes lose their filopods and become rounded, indicating cytotoxic activity of the bacteria. To quantify this cytotoxicity, several flow cytometry tests such as adherence test and phagocytosis test, and reactive oxygen species (ROS) were used. To compare the cytotoxicity degree, 2 bacteria strains of V. tapetis that pathogenic to bivalves and the mutant bacteria were tested. The results of this study showed that V. splendidus, showed lower cytotoxicity degree compared to Vibrio tapetis as positive control and comparet to the normal control.
