PENGUJIAN EFEKTIVITAS EKSTRAK DAUN KELOR DALAM PENGENCER SITRAT-KUNING TELUR TERHADAP KUALITAS SEMEN CAIR BABI LANDRACE

Abstract

The aims of the present study were to evaluate the effectiveness and the best concentration of moringa leaf extracts (MLE)in Citrate-egg yolk (CEY) diluent on the quality of liquid semen of landrace boar. The semen samples were collected twice weekly from three landrace boars (2-3 years old) in normal reproductive performance following the massage method. The good quality sample was divided into five tubes, each diluted with CEY (P0), CEY + MLE 5% (P1), CEY + MLE 12.5% ​​(P2), CEY + MLE 22.5% (P3) and CEY + MLE  35% (P4)following a completely randomized design consisting of five treatments and four replications. The diluted samples then stored in a styrofoam at a temperature of 18-20˚C and were evaluated for motility, viability, plasma membrane abnormalities as well as the integrity of every 4 hours. The results showed that the spermatozoa kept during 24 hours in a diluent of CEY + MLE 5% (P1) had higher (P <0.05)motility (42 ± 7.58%) and viability (63.62 ± 13.24%)  than the other three diluents. However, there were no significant different (P> 0.05) between treatments forthe abnormalities and plasma membrane integrity of spermatozoa. Adding moringa leaf extracts (MLE)intoCitrate-egg yolk (CEY) is effectively maintaining the quality of liquid semen of landrace boarat the best concentration of 5% moringa leaf extract (MLE)

ABSTRAK

 Tujuan penelitian ini adalah untuk mengetahui efektivitas dan konsentrasi terbaik ekstrak daun kelor (EDK) dalam pengencer Sitrat-Kuning telur (SKT) terhadap kualitas semen cair babi landrace.  Semen dikoleksi dua kali seminggu menggunakan metode masase dari tiga ekor babi jantan landrace berumur 3 tahun dengan kodisi organ reproduksi yang normal. Semen yang menunjukkan kualitas yang baik  dibagi  empat, masing-masing diencerkan dengan SKT (P0), SKT + EDK  5% (P1), SKT + EDK  12,5% (P2), SKT + EKD 22,5% (P3) dan SKT + EDK 35% (P4). Semen yang telah diencerkan disimpan dalam stirofoam pada suhu 18-20oC dan dievaluasi motilitas, viabilitas, abnormalitas serta keutuhan membran plasma sperma setiap 4 jam.  Rancangan yang digunakan adalah rancangan acak lengkap yang terdiri atas lima perlakuan dan empat ulangan. Hasil penelitian menunjukkan bahwa spermatozoa yang disimpan selama 24 jam dalam pengencer SKT + EDK 5% (P1) memiliki motilitas 42±7,58% dan viabilitas 63,62±13,24% lebih tinggi (P<0,05) daripada ketiga pengencer lainnya, sedangkan abnormalitas dan keutuhan membran plasma spermatozoa dalam keempat pengencer berbeda tidak nyata (P>0,05). Kesimpulan dari penelitian ini adalah EDK yang ditambahkan ke dalam pengencer SKT efektif dalam mempertahankan kualitas semen cair babi landrace dengan konsentrasi ekstrak daun kelor terbaik adalah 5%.
